PHARMACOLOGICAL ACTION
Amoxicillin trihydrate is an antibiotic that belongs to the group of semisynthetic penicillins (aminopenicillins). The mode of action of the drug is inhibiting the synthesis of cell membrane mucopolysaccharides this resulting in their impaired barrier function and the occurrence of an osmotically unstable spheroplast. The exact mechanism of this effect is yet studied but amoxicillin is known to be bound with several ferments (carboxypeptidase, transpeptidase, edopeptidase) inside the cytoplasmic membrane of bacteria that synthesizes the cell wall.
The drug is active against gram-positive bacteria (Actinomycos spp., Bacillus antracis, Cloctridium spp., Corynebacterium spp., Erysipelothrix rhusiopathiae, Streptococcus spp., and Listeria monocytogenes) and gram-negative bacteria (Actinobacillus app., Bordetella bronchiseptica, Fusobacterium spp., Haemophilus app., Moraxella spp., Pasteurella app., Proteus mirabilis, and Salmonella spp.). Insensitive to the drug are such microorganisms as Pseudomonas aeruginosa, Serratia, indole-positive Proteus (except for Proteus mirabilis), Enterobacter, Citrobacter and Acinetobacter. The drug is also inactive against Rickettsia, mycobacteria, fungi, mycoplasmas and viruses.
Prednisolone influences actively on carbohydrate metabolism, protein metabolism, water metabolism and salt metabolism. It contributes to glycogen accumulation in the liver, increase a blood glucose level, induces increase in nitrogen elimination with urine.
Prednisolone manifests anti-inflammatory, desensitizing, antitumor and antiallergic actions. Also, it has antishock and antitoxic actions.
Prednisolone has been reported to decelerate the release of cytokinins (interleukins 1 and 2 and γ-interferon) out of lymphocytes and macrophages, to inhibit the release of inflammation mediators by eosinophils, to reduce arachidonic acid metabolism. Stimulating steroid receptors, it induces the formation of a special class of proteins – lipocortines, which have an antitumor action.
USES
The drug is used to treat subclinical and clinical forms of a bacterial etiology mastitis caused by microorganisms sensitive to the drug ingredients in cattle, sheep and goats.
DOSAGE AND ADMINISTRATION
Cattle:
Ѕ to 1 syrette depending on disease severity or 5 to 10 mL of the suspension per each udder quarter, intracisternally, once daily.
Small cattle:
ј to Ѕ syrette depending on disease severity or 2.5 to 5 mL of the suspension per each udder quarter, intracisternally, once daily.
Treat for 5 to 7 days. Prior administration, milk dry an affected udder quarter.
CONTRAINDICATIONS
Hypersensitivity to drugs belonging to the penicilline group.
The drug should not be combined with antibiotics of the cephalosporin, tetracycline and macrolide groups.
PRECAUTIONS
Animals may only be slaughtered for meat and milk is fit for human food in 14 days and 4 days respectively of the last use of the drug. Until this date, meat and milk is fed to non-food-producing animals or disposed (according to veterinarian’s conclusions).
